5 minute animation on a _topic_? Personally, I like stories over topics any day. You could do a short animation of a fairy tale or folk tale, or you could come up with a unique idea for your own story.
When I'm trying to come up with an idea for something like this, I'll sit down and write out a hundred ideas (good or bad), then pick a few that I like and develope them a bit. Then I'll pick the best out of those and finish it off.
The key to coming up with good ideas is to create a lot of ideas and then pick the best from them.

Keep in mind 5 minutes is a LONG time for an animation, even if you take a low framerate. Do the math; 5 (minutes) x 60 (seconds) x 12 (random frames per second) = 3600 drawings. Even if you go 6fps, you'll need 1800 frames :)
Anyhow, provided that you have the time and drive, pick something that is really really reaaaally close to your heart. It'll be an incentive for you to finish and keep on trucking!
You could do a little story of a character ? The journey of a leaf blowing in autumn ? The odyssey of a dandelion seed in Spring ? Someone mouthing something, and you need to read on their lips ? A Cadavre Exquis kind of animation, having one shape transform into another, whether they're representative or abstract?
etc.

A video of people counting 300 seconds - with perfect lipsync.
A video of people from around the world saying "five minutes" in different languages, again perfect lipsync.
What else is a five minute thing? You could do a really detailed vid about making and enjoying a cup of tea, and all the little things that you can see at the same time, like the steam rising, fingers twiddling, the tea "ink" seeping into a cup.
Or someone running a 1 mile race, and what goes through their mind in that 5 mins.
Or animate a variety of clocks running through the five minutes (digital, analogue, go through the workings of a clock)

A suggestion might be to show "the life" of something over a period of time. What happens to a tree, or a dog, or a car, or anything, over say, a 24 hour period. Or really fast over the four seasons of the year.
Eg., for a tree -- people might read under it, put a swing on it, carving a heart into it, etc. if you want a sad ending it could be chopped down.

I like the 5 minute thing, why not do it on that 5 minutes of brainstorming about putting your animation together, but make it humorus.
