If you are going to be using the train a lot, you should get a JR Pass. 28,300 yen (US$242) buys you unlimited access to all JR trains everywhere in Japan for seven days (except the Nozomi shinkansen, but all the normal shinkansen lines are OK). You can also get a seat reservation on any train for free. If you plan to travel long distances within one week, you will save tons of money.
To use this pass, you must be a foreigner with a tourist visa. You also must buy the voucher for the pass OUTSIDE of Japan. You can't buy them in Japan. Once you arrive in Japan, you can activate the voucher and get the pass at many JR stations, including at Narita airport station.

Taking the fastest trains available will cost you ¥18,000 or so. That's Narita Express to Tokyo, then Nozomi to Osaka. About 4 hours total.
The cheapest alternative, if you aren't in a hurry, is to take the Keisei limited express train to Nippori (¥1,000ish), Yamanote Line to Tokyo Station (¥150ish), then an overnight bus to Osaka (¥4,000 to ¥8,000 depending on how much comfort you want). You'd get to Osaka around 7AM the next morning.

Hmmm. My source is telling me it is only 8720 yen. This source allows you to book a reserved seat, which equates to what the other posters have said. I would advise not booking the seat. It's not worth the extra 6000 yen. The Shinkansen isn't very busy, and if you travel on normal days, you'll be able to get a seat.
The only way I would be wrong is if the 'Reserved Seat' is always part of the price. But if it was, then why wouldn't it just be part of it and not a separate entity. Plus I heard one week passes for foreigners on Shinkansen is 10000 yen, so you may as well buy that if that is true.
