Isn't it they said it should be the blood of the son of Bootstrap Bill, which is Will Turner? Why did Jack Sparrow use his blood in the end? What's the twist of the story? This is from the Curse of the Black Pearl.

Will Turner's case was separate. Everyone had stolen the gold except for Jack who was stranded on the island after they mutinied.

In the scene where he's laying out the plan for Captain Barbosa, he steals one of the coins by taking it out of the chest inheriting the curse on everyone else. He needed it in order to buy enough time till Will could lift the curse by getting to the treasure chest and putting his own blood in.

While Captain Barbosa was cursed, he was invincible. The pistol that Jack had would be useless until the curse was lifted. However, since Jack had to buy time until "the opportune moment," he had to even up the score by stealing a coin so that that he'd be temporarily immortal as well while fighting Captain Barbosa as an immortal.

That's why when Barbosa drives the sword into Jack Sparrow, Jack falls back into the moonlight revealing his cursed nature. He then whips out the coin with his skeletal finger flicking it back and forth with a sly grin and says, "I couldn't resist, mate."

Barbosa thought he had the advantage, but Jack had outsmarted Barbosa revealing how he'd evened the playing field. Jack was being sneaky.

If Jack Sparrow tried to hold off Captain Barbosa as a mortal, he'd be getting all knicked and cut up or even get mortally wounded without the curse serving as a shield. He was fighting fire with fire by challenging Captain Barbosa in the immortal realm.

That's why Captain Barbosa in mid-fight asks if he and Jack are just going to be two immortals locked in combat until judgment day and trumpets sound.

When Will finally got into position to remove the curse with his blood, Jack cuts his own hand bleeding on the coin he stole and throws it to Will who adds his own blood before dropping it back into the chest removing the curse altogether.

That's when Jack shoots a hole in Captain Barbosa just before Will drops the remaining coins in so that the curse is lifted allowing Captain Barbosa to be mortal again. In doing so, the curse was lifted right when Barbosa was mortally wounded witha shot to the chest.

It is Will's blood used. Will cuts his own hand open and drops the coins in the chest, thereby reversing the curse. Jack's blood had nothing to do with it.
