I have seen this happen on a computer on which someone had recently plugged in a USB sound device. Their previous sound card no longer worked once the USB sound device had been connected.
The solution was to go to the Control Panel and open the Sounds and Audio Devices control panel. Then click the Audio tab (at the top), and make sure the correct sound card is selected in each of the categories. Do the same for the Voice tab.

Try using different speakers or just the mp3 player headset in the socket to rule this out then try opening everything that plays a sound and check it hasn't been muted a bad peice of software can mute everything.

If your mutes are on, the drivers properly installed, speakers not blown the only thing left is the sound card. Remove it and take to pc repair facility and have it tested.

I have a similar problem with a Toshiba Satellite A35 laptop. All software audio diagnostics say the sound is fine, but it quits working. The computer is currently in the shop for troubleshooting.

Try to restart the computer, turn it back on. Restarting the computer a few times may make the sound start working again.

Did you check to see that your speakers are plugged in? Is the sound on "mute"?
