You have not stated your departure point. If you are in America fly to London first, it will be cheaper, then get EASYJET flights to Rome and back to London (easyjet has the cheapest flights in Europe.)
If you are in the UK, go to London by train, and still go easyjet to Rome...the fastest way to travel is by plane... Otherwise the Eurostar will get you to Paris, stop there for a couple of days, then go on to Rome by train...Don't sleep in the train, if you are in a compartment with other people, (up to 6 persons) you won't sleep comfortably, and some have had their belongings stolen ...and if you are in a private compartment for the 2 of you, it could as expensive as a good hotel room!
Both cities are magnificent to visit... In London you have the day passes on the doubledecker buses (a great tour system, you get on and off the buses wherever you want to and visit everything at your own speed and leisure, the tickets are valid 24 hours). If you live in the UK, you should have other chances to visit London, so spend more time on the continent... If you come from other countries, US or Europe, then share time out equally between both..
Rome may have something similar to the day pass, but I'm not sure. .
In Rome the Vatican museums are magnificent, as are all the ancient monuments...plus the city itself is worth jast walking around in to see things... Piazza di Spagna, Piazza Navona, Pantheon, Colisseum, etc.
In London the royal palaces (go to Windsor also, it is just outside of London), the museums are also excellent. Depending what time of year you are going, the weather will probably be better in Rome. So you might want to take a raincoat for London...
In London you can also enjoy an evening at the thater and some night life. "Pub grub" is fun!
And italian food is wonderful, enjoy restaurants in Rome.And do some "people watching"!
