area of a room = height x width x length
area of a balloon = (roughly) 4 x 3.14 x radius x radius (radius is the width of the balloon, filled, divided by 2)

you could to the math and then divide the area of the balloon into the area of the room, and that will give you a rough guideline.... or you could just blow up a LOT of them and take your chances.

I'd do the math, but I'm like that... have fun!! :-)
