It depends on how you define that. USA and Russia, along with China, are the only nations that have used their own spacecraft to send people into space. However, other nations do have space programs, and have sent people into space using the equipment of other countries. These nations include France and Canada.

Strictly speaking - US and USSR (Russia) and China.
But really, neither the US nor the former USSR did it all by themselves. They both grabbed German scientists after WW II.
Then Canada played a huge part in NASA from day one, sending thousands of the world's best scientists and engineers south in 1959 to help found the company.
And if things go according to plan, there will be a launching pad in Nova Scotia to launch commercial space ventures in a few years.
