Yes. Your will can state whatever you want it to without regard to marriage, separation, divorce, etc. Remember however, if you merely divorce and you live in a state with community property laws, then the wife is entitled to anything jointly held. Obviously, if you divorce, you would still be alive, and thus your will would play no role at that time.

This all depends on the state. In a community property state, half of everything is hers. However - there are exceptions. If it's a second marriage, it can be more complicated. The wife should see an attorney to know for sure.

It depends on what you mean. He can leave his personal property, and his portion of the marital assets to anyone he wants. That's how it is.

If you own a house together, most states allow the surviving spouse to stay in the house until death.

You can try, but she'll probably fight it in court and win half anyway. They figure if she was married to you all that time she deserves some compensation.
