The closest big casino to San Francisco is River Rock in the Wine Country (you can combine it with a visit to the wineries):

The largest casino in Northern California is Cache Creek near Sacramento. It's further away, but you'll get more amenties than River Rock (like floor shows and buffets).

There are some casinos closer to the city... in San Pablo and San Jose. But as others have pointed out, they're a little depressing: small, hardcore gamblers, and no comps.
California's so-called "ban" on gambling is something of a joke. The state now allows corporations to buy up land and build casinos, even if the "tribe" they supposedly represent has never been anywhere near the area. Palm Springs, in particular, has become the California version of Reno.

The closest "Vegas" style casinos are located in the Sacramento area.
There is Cache Creek in the town of Brooks which is in Yolo county north of Davis. Then there is Thunder Valley off of Hiway 65 near Roseville. Lastly there is Jackson Rancheria in Jackson, about an hour east of Sacramento in Amador county south of Placerville. All are at least a 2 hour drive from San Francisco, more on a Friday.
There are "casinos" in and around the San Francisco area, most notably in San Pablo, but these are nothing more than Poker rooms.
Good luck!

Las Vegas style casinos are illegal in California. The exception being those facilities that are situated on "Indian land"
There are several in the Sacramento area..approx 90 minutes outside of San Francisco
The largest one is Cache Creek. They have a hotel, spa, and other amenities.

If you just want to gamble you can go to a card room like the oaks in emeryville
http://www.oakscardclub.com/
There is a casino in San Pablo (north of Richmond, 20 minutes from SF):

Hopefully you won't be donating too much money to the latter, they're already trying to expand and traffic on 80 is bad enough as it is.

Ok, so I didn't know there were casinos in SF. I thought the only way to gamble in CA is on a reservation. It is weird though because you see exceptions around and wonder how they get away with it!

Lucky Chances card room as was mentioned is closest, but that's no way to gamble. Best head up for Tahoe. Only a 4 hour ride and really beautiful.
