I would ask what my specific duties would include and if there exists a job description. I also would want to know the chain of authority in the organization and specifically the person to whom I would be responsible. Then, of course, I would ask about fringe benefits and salary. Most important of all, I would ask what the opportunities for advancement are in the company.

How did this position become available?
What is the average amount of time a person stays at this job?
Is there room for growth?
do you have any concerns about my ability to do the job?
How would you describe the management style here?
How do you see this job changing in the next 5 years?
What are the Departments long and short term goals?
What is the schedule like? is it flexible?
What problems if any have employees encountered doing this job.?

Dont ask about pay/benefits/vacation until you have been offered a position.. it make it look like this is all you are interested in.

How about:

What was it about my resume/application that suggested you call me in?

Do you expect growth in (staff, production, sales, locations) ?

Where would I fit in the organization?

How large a staff in my immediate (department, location, function) ?

May I meet my potential supervisor and co workers?

(whatever you ask, show some curiosity and enthusiasm about the job.)
