Shaun Alexander, Larry Johnson or LaDanian Tomlinson. Shaun Alexander is a little scary because, a) he's on the cover of Madden 07 and b) his team lost the Super Bowl last year. But who believes in curses? Johnson closed out the season incredibly well after Priest Holmes went down last year, and he's got a shot at the rushing title this year, but he is young and hasn't demonstrated his consistency as a fantasy back over multiple seasons yet. Tomlinson has been consistent over several years, misses very few games, and should be a relatively safe bet. I would probably pick him 1st.
As for Carson Palmer, or any other quarterback, don't pick them until at least - AT LEAST - the 4th round. The difference in points between Palmer or one of the Mannings and Trent Green or Kurt Warner or another back you can get in the 4th round is nowhere near as big as if you don't have a premier back.
You're going to have to be as concerned with your 2nd & 3rd picks as with the 1st. Assuming you are using the "snake" type draft order, once you have picked one of the top 3 running backs, you won't pick again until picks 20 and 21. It's possible, depending on who is left and the league format, that you might want to pick 2 more rb's, but you will probably want to go with an RB and a WR. If you can't get an every down back, go for the big back who scores td's. (I'd say someone like TJ Duckett, but who knows what role he will play in DC).
Plan your draft so that you are ready with both picks each time. Unless you get lucky and somebody drops into your lap, you will probably be looking at a lower tier of talent each time your 2 picks come up. If someone you really want is on the board when your picks come up, you better pick him because he probably won't be there 19 picks later.

1 guy alone will not let you dominate anything. That said probably your best week in and week out point producers would be Larry Johnson, Shaun Alexander, Tomlinson, and probably Peyton Manning. I'm in 6 leagues and those 4 have been the top picks in 5 of the 6, with good reason.

(haha) at the dude that said carson palmer for the first pick of the first round. If you draft first go with Sean Alexander. He is a big, durable, and most importantly consistent back. People will say draft Larry Johnson, Larry Johnson this, LJ that...but to be honest he only had One great year. It was a monster year true, but still you can't judge consistency off of one year. I would probably draft tomlinson before LJ just to be safe, but that's a pick that the masses would shun. So all in all...draft Alexander.

Tiki Barber. The Giants have ALL their starting O linemen returning this year to block for the underrated back. If not Tiki go with Larry Johnson (decent O line) Warrick Dunn (Less decent) or Shaun Alexander (although I'm superstitious so I wouldn't pick him)
