No, and its not really a nessisary test. First you get the blood test if it comes back saying there is a potential problem with the baby (which is 80% a false possitive that something is wrong) then they send you for the Amnio Test which has a 5% chance of hitting the baby and causing a misscarrige.

Yes. They numb the area before they do the extraction of fluids through your navel. It doesn't hurt while they are doing it, but I was a little sore afterwords. It went away in a couple of hours.
It's like having blood drawn, it's a slight prick then the area is tender.

Yes. They use a long needle and stick it through your bellybutton. They take amniotic fluid and that is it. They use a bit of lignocain to remove the "feeling". It feels uncomfortable, but is not really sore.
