This question can be answered in multiple ways and I will try to do so:
Movies (year of release in brackets) that had a very long run in theatres:
First was Alam Ara (1931), then came Kismat (1943) which ran for 5 years before a fire destroyed the theatre in which it was running, then came Sholay(1975) which was removed from the theatre in which it ran for 5 years due to the release of Shaan which had the same team. The current record holder is DDLJ (1995) which has run for 10+ years in Mumbai. This record does not seem to be broken given today's dynamics where super successful movies dont run for more than a couple of months.
Movies (year of release in brackets) that set the records for the highest grossers ever:
First was Kismat(1943), then Aan (1951), then Mughal-E-Azam(1960), Sholay(1975), Hum Aapke Hain Koun (1994) and then the currently record holder Gadar Ek Prem Katha (2001)
If you take inflation and rise in population then the most successful and popular movie of all time is the incomparable "Sholay".

SHOLAY- It ran at a Bombay theatre (I forget the name, Maratha Mandir or Liberty) for 5 years, the first three were booked everyday on advanced tickets.
