If you could be more specific, i will be able to help you. We can create two modes of layouts. 1. Using tables
2. Using div tag (CSS)
I suggest you use tables to create your HTML layout as div is a bit complicated. what we designers do is , we create the design in 779X600 size in photoshop and then we slice it and creat the HTML using the dreamweaver software. Dreamweaver is a bit costly. hope you try this out.

instead of making html lay out through manually (writing all the codes) try some new softweres which makes ur work so easy and impressive, go and get like front page and dream weaver
