It depends on what mail client you are using.
In Outlook, you do this:
Click Tools, Options
On the Preferences tab, click Email Options.
Click Advanced Email Options.
Under "When new items arrive in my Inbox", you can see 4 different kinds of alerts,
- Play a Sound
- Briefly Change the Mouse Cursor
- Display an Envelope in the Notification Area
- Display a Desktop Alert.
Whichever mail client you use should hopefully also have such options - use the Help function to find out where to configure them.

if your using Outlook Express, which I presume you are, leave it minimized and go to:
XP: Start-control panel-sounds, speech and audio devices-change the sound scheme-New mail notification. Then assign a sound to it.
98, ME, 2000:Start-settings-control panel-sounds New mail notification. Then assign a sound to it.
Don't quote me on the 98/ME/2000 I was just doing it off the top of my head and its been a while.
Good Luck. :)
