First you can clean-up the disk. When you do so, some files written as fragments may br left on the disk. Once you have defrag, there will be spaces left so that you can defrag the disk.
So I would rather clean the disc as frequently as possible (once a day) and defrag the disk when the computer slows down.

You should do Disk CleanUp, then, Check Disk, then Defrag. You should also try out that System Mechanic 6 Program, I've heard good things about it.

disk clean up first then defragment, that way it can open up any sectors then the defrag can close them.
Please dont use microsoft's defrag its sucks!! use perfect disk 7.0!!
something else u can do, is try registry mechanic
other things, run weekle virus and spyware checks, dont load up a lot of programs when windows stars and finally increase ur ram!!!

I personally run two antivirus software programs, then clear my cookies and browser history, then disk cleanup, then defragment.

Doesn't matter, but cleanup is a lot faster and you might want to get it out of the way first.
Hope that helps.
