Physically opening a router is a bad idea. There are no user-serviceable parts. You'll just ruin the router. A sledgehammer would work as well as a screwdriver.
If you're talking about accessing the router to do configuration, it depends on the type of router. For some, you have to telnet in. (From a command prompt, type telnet xxx.xxx.xxx.xxx (xxx is the router's IP address). With the username/password, you'll access the configuration screen. If your router has a web-based interface, you can access it by opening internet explorer, and typing http://xxx.xxx.xxx.xxx. Once again, with the username/password, you'll get a configuration screen.

With a screwdriver usually, but there isn't anything in there most folks can do anything with.

Commercial grade equipment would allow you access to upgrade memory, but that's about it.
