Your first two picks should be Running Backs followed by a strong, consistent QB. I would recommend either Shaun Alexander, Larry Johnson or LaDainian Tomlinson as your first pick. You can't go wrong with those guys.

While Manning would be your first choice if you were a real live "GM" for an NFL team, in fantasy the written rule is to draft a stud RB in the first round, and usually first 2 rounds.
You cannot go wrong drafting a Larry Johnson, Shaun Alexander or LT with your first pick. What you need to prepare for is not the first round but the later rounds. These are the rounds that seperate the winners from the losers.

Definitely pick an RB. They are your workhorse. They get the most points week in & week out. (In the first round, only Peyton Manning should be considered & even he isn't a top three candidate this year.)
Of the top three fantasy RBs (Ladanian Tomlinson, Shawn Alexander and Larry Johnson), I lean towards Shawn Alexander. All three are great RBs, but Johnson just lost his starting left tackle to retirement (Willie Roaf) & LT has tougher defenses in his division than Alexander does. I don't think you could go wrong with any of the three, but I'd suggest Alexander, then Tomlinson, then Johnson (in that order.)
In the second round, you might want to consider getting another RB, depending on who is available & how many RBs you have to start every week. But if QB Tom Brady, QB Matt Hasselbeck or WR Steve Smith are available, they're decent options in the 2nd round.

Without knowing your scoring system and such, I would still advise you to pick a running back. I like LT. Larry Johnson will have trouble with his line and the loss of his fullback.
