How big is a grain of sand in a universe once full of nothingness?

Can a universe ever actualy be full of nothing or something or does it need a mix of both to exist? Say you take a box and make a true vacuum in it can that ever even be done? Is it possible to make a spark of life inside that box and have it grow forever?

A grain of sand will still be very very small even in a universe full of nothing.
