Your speakers typically have a built in amplifier, the AUX IN is just another input to that, which you can use.
In other words you could hook up an ipod to it or some other source of audio, and have it come out the speakers also.

AUX means Auxillary, or sound from another source.
Its a place to plug in something else, like a CD player or IPod, to play music over your speakers.

Vista is a little cranky when you switch from speakers to headphones and back. I see you've already rebooted. Hmm.... OK. Put the green cable back in it's correct slot, make sure the Realtek HD Audio Manager options are set correctly and shut down the PC. Leave it off for about 30 seconds for the memory to fully clear. Boot it back up and see if it works. If it doesn't, contact me and let me know.

Its a signal input port, in other word it brings an audio signal from a computer or stereo and is heard from the speakers, kind of like the audio in jack on the back (or front) of your VCR. or pugging in a set of headphones into your portable cd players.

AUX IN - Auxilery In - It is used if you have a fancy sound card that can do true surround sound, dont worry about it and just plug it into the green hole!! Good Luck

It's a second input incase you wanted to hook up a different device like CD or MP3 player.
This spot is NOT for headphones. NOT for headphones. If you do put headphones in this slot you will be sorely dissapointed.

AUXiliary INput is used for plugging in third party devices (mp3 players,FM radios etc.) directly to your speakers. I presume that those are active speakers so you can use them even when your computer is turned off.

For an external audio source such as ipod, walkman etc.
Not for headphones, the speakers and headphones are both output devices
