There are 3-4 steps in the making of a functional protein
First, the putting together of the amino acids. Then the folding/splicing into a shape. There may be additional molecules added to give a different charge. Next is putting that together with other protein chains, like hemoglobin.
It is the final product that the antibody responds to. Kind of like reading a sentence. I can respond to your question, but if I untangle all the letters, and put them in order of the alphabet, I can't read it

Because denatured proteins are a different shape than their native forms so the antibody binding site will appear different. It is like taking a hammer to your front door key and asking why it it fails to open the door anymore.
