If were talking about a regular PCI slot video card then the answer would be yes. The problem would be if you plan on doing some HARDCORE gaming you're going to run into some problems because the response will slower than that of the AGP card.
If you're overall plan is just to multi task then you'll be fine.

Save yourself some headaches by making sure both videocards are made by the same manufacturer.

There is no way to mix 2 different types of video cards at the same time. Unless you are doing an SLI/Cross-fire setup with the correct bridging with 2 identical video cards.

If you mean can you run PCI-Express card in an AGP slot, then the answer is no.
