Hard to tell why the animosity is there. Maybe jealousy. Or maybe the Assistant Manager feels intimidating by you whether you are or aren't trying to be that way. I know women who get this way when the new hire is a beautiful, outgoing, intelligent, young woman. Does this fit you?
You can quit any time you like. Don't let a drug store employee tell you what to do.
You can email me if you want a clearer choice. I could help you decide better. I've had jobs I knew I would hate, and have walked out on, only after a few hours!
No job, unless you sign a contract, can make you stay. You take a job on your terms only!

Starting a new job is always hard, especially when it's your first. My advice, give it another chance. Go in there telling yourself that you won't let her get you down, she's treating you this way because you're new, it happens all the time. After a week or two, if nothing changes, then there is absolutely nothing wrong with quitting. Never do something that makes you unhappy. It would be common courtesy to give at least a week's notice and it would be wise to find another job before you quit, but if it makes you feel that bad then leave. You only live once, don't waste it on doing something you hate and making yourself miserable. Good luck :)

Talk to the manager about how you're being treated. At any rate, don't work under those conditions! If you're not comfortable with talking to the manager and having to go back to work under the assistant manager, then quit but at least report how you've been treated and let the manager know why you're quitting. I'd even go a step further and put in writing to both the manager and whomever the manager reports to how you've been treated, particularly if you're doing a decent job and this animosity toward you is not because of something you've done. Make sure you get a copy to the head of the ladder -- they need to know what kind of people they have running their business. Be sure and collect any pay they owe you. Life's too short to be that miserable with any kind of job.

Don't let anyone run you off of a job. Learn to do say "so what". So what if she doesn't like you if you know that you are doing your job well forget her, the peopel who really matter will start to notice. You have only been there three days and of course someone has to be the target. If it doesn't get any better after two weeks go over her head. Explain to her boss that you like your job and you are thankful for the opportunity, but you will not be but in an uncomfortable position. In the mean time keep records of everything that is going on.

You can quit whenever you want to.
If you quit without giving a notice, you won't get a good reference from them, however. So, on your next job application, don't mention the drug store.
Before you quit, sit down and be honest with yourself. If the situation is TRULY as bad as you've described here, then you should quit. Are you being overly sensitive? Why would the assistant manager hate you? Is it her or is it you? Is there someone else at the store you can speak to? Is that job better than flipping burgers?

The assistant manager is a jerk and knows nothing about managing. Ignore this person. What the hell does asst manager mean anyway? Nothing!
Tell the real manager how you feel and that you intend to quit over this treatment immediately if it is not fixed. He or she will fix it or you will leave. It's that simple.
You didn't hire on to be abused; don't put up with it. In five years it will be forgotten.

Life is more than a 3-day job. If it is causing you this much grief, even in such a short time, then it's probably time for you to get out and find yourself a job where you will be happier.
Plenty of other companies have jobs that might be right for you -- better environment, better people to work for and with, better job.

We had a brand new electrical engineer that left after two days on the job. He will most likely never work for my company again, but that was his decision to leave. Just remember that by quitting after 3 days means that you have burnt that bridge.

the first few days are hard..you need to tough ti out for at lest 6 months Hang in and good luck also speak to the store manager if you feel the you are getting a hard time
