Over the course of my career I have hired hundreds of people, fired dozens, and worked with thousands. I don't believe that I have ever hired anyone who didn't send a thank you note of some kind - at least an e-mail - after the interview. A printed letter mailed in a business-sized envelope is still best in my view.
The best letters say something like this:
"Thank you for taking the time to meet with me today about (the position). I enjoyed our conversation, and as we discussed I believe my (specific attitude, bearing and skill that is relevant to the job) make me an excellent candidate. When we talked about (subject discussed during the interview) I felt that we were closely aligned in our thinking, and I am looking forward to further conversations.
I will contact you again in (about ten days) to follow up further.
Once again, thank you for your time and consideration.
Regards,
Your signature.

Interviewers appreciate "Thank You Letter" It shows that you're gracious and appreciative. Also it will make you stand out in hundreds of job applicants. You could either email it or send it via snail mail. I usually do both. It works like a charm. It will get you at least to the 2nd job interview or the job offer.
This one is simple no BS thank you letter. It got me a job with $20K raise!

Actually, if it's possible, you should have a thank you letter ready - after the interview, you'd drop it in the mail ASAP so that the interviewer receives it ASAP. When in pre-interview mode, you should get the interviewer's name and title (and proper spelling).
There are many templates around - try the following links:

Yes. Simply because it's an opportunity to job their memory. I would write something like: Thank you for taking time out of your busy schedule to meet with me. (You're showing him that your appreciate his time and know it's valuable). Since then I have been doing some research on your company/corporation. I am very enthusiastic about joining your team. Awaiting your response, I remain, sincerely yours.....sign your name.
