You can't physically use Hard disk space as RAM or system memory. Instead you can use it as Virtual Memory. To do so, follow these instructions:
1. Right click on My Computer, then click on Properties
2. Click on the Advance tab
3. Click on Settings under Performance
4. Click on Advance tab
5. On the bottom where it says Virtual Memory, click Change.
6. Increase to your desire.
Note: Typically, the system allocates an exact amount of your physical RAM plus a half... For example, if your PC has 512 MB of memory, your system would assign 768 MB of Virtual Memory.

I believe you are talking about virtual memory. Instead of using RAM, the computer will use the harddrive to store some data for the running processes.
Virtual memory is much slower than actual RAM. You can adjust the amount of virtual memory you want to use by Right clicking My Computer > Properties > Advanced and then adjust the virtual memory settings.
It is best to leave it at defualt setting so make sure you know what you are doing before you make changes.

Yes, you can use hard-drive as RAM, but it has its own limitation, it is not that you can assign any space and that will be utilized as RAM, however here are the steps you will have to manage Virtual Memory to do this:
Go to System properties >> Advanced Tab >> Virtual memory.
