Transmission fluid is to be checked HOT by design. Engine oil on the other hand can be checked both hot and cold, as it is common for the engine to use oil as a part of the engine lubrication design.
There will be some slight differences between hot and cold, but they are very slight, and built into the marks of the dipstick and crankcase design.

doesnt really matter if it is hot or cold just as long as engine has been sitting off for at least a 15 minutes or so to get best reading. That way all the oil from the top of the engine has had time to drain down.

When the vehicle is hot, because the vehicle burne the oil when it has started or when u drive it.

PREFERABLY WHEN THE VEHICLE IS COLD, THE HOT OLD WILL RISE ON THE DIP STICK CAUSING A FALSE READING, ON THE OTHER HAND WHEN CHECKING THE TRANSMISSION FLUID YOU MUST DO THIS WHILE THE CAR IS IDLING
