You ought to be country specific.

The first ones I know of in this country were made in the early 50s by the Us Government and shown in public schools as part of a program to prepare Children for a possible nuclear attack be the Soviets.

The Soviet government also did some animated films along the same vein as America the possible enemy.

There was an animated film made in Japan about the same time. It was a short, about fifteen minutes long.

My personal guess on this would be the Gojira movie from 1954, although we know it today as the first Godzilla movie with the Raymond Burr footage added for American audiences.
For a strictly TV answer, I would have to say it is what we know as Astro Boy here in America.
