Be sure to check out Manga - Sixty Years Of Japanese Comics by Paul Gravett. It's ISBN is 1856693910. It's one of the best guides to the history of manga in Japan. It's a good mix of intelligent analysis, history and (appropriately) it's full of pictures.
No anime fan worth his salt has not seen all of the following - Akira, Ghost In The Shell (and its sequel, AND it's tv series companion Stand Alone Complex), and Ninja Scroll.
One of the strengths of manga and anime is the sheer volume of stuff available in translation - a huge percentage of Japan's printed output is manga, and their animated adaptations appear this side if the world pretty quickly.
Forbidden Planet always stock a wide range of translated manga (and flipped - reading order is reversed in Japan). Most good bookshops have realised the popularity of the genre too, so even a bog-standard Waterstones usually has a selection (although it can be pretty poor). Stick with Forbidden Planet. And no, I don't work there or anything, I just spend half my wages on them.

Check AnimeNfo.com...that can give you titles of anime and manga that might suit your above requirements. You can do searchs by genre.

For example, the horror genre would cover vampires, etc., mecha would cover robots, hentai would cover erotica, romance is obvious. .

Some titles that might work for your brother are Hellsing, Witch Hunter Robin, Neon Genesis Evangelion, Mai Hime, Elfen Lied, and Rahxephon.

DVD or comic version?

Tell your brother to key in anime into amazon and he will get a lot of results. Wikipedia is also good for this. If he tries the magic words "hentai" or "erotic manga" on Amazon he should find such "gems" as ultra gash inferno or beauty labyrinth of razors ...

Your brother should find a lot of material to assist with his research, but he should also remember the context of anime and manga as they are not just viewed as comic books in Japan and are targeted at different markets. He could also look at how manga and anime are reflected in PS2 and other video games.

Now, my toddler wants to watch "Spirited Away" which is a superb anime cartoon which features mystery, demons, ghosts, witches and a young girl gaining maturity. Well, it's as good a place to start as any :)

You can check out Tokyopop.com for mangas that suit your tastes.

I think there is a manga called " The Vampire Game" and it has a vampire reincarnated as a cat to get revenge on a king who killed him.

Anime: Elven Lied.
It doesn't have vampires...
And not really witches and robots...

It does have mystery, demons, erotica (not much but I think it has some...), romance, violence.

Urotsikodi or soemthing liek that. Subtitled legend of the overfiend. Not too sure abotu robots but it has most the other stuff, Terntacle sex too in parts.
