1)Is overlap of genes allowed? Are any regions excluded as promoters and regulatory regions? I guess the answer is no to both so you simply have to do the math yourself. Just a hint. The avearge weight of amino acid is considered 135 daltons... And 4,000 daltons is actually a very small gene (approximately 29 amino acids)...not an average. Probably you mean 40,000...

2)DNA denatures. Single stranded DNA can absorb better light at 260nm than double stranded DNA. The maximum difference is approximately 36% of the initial value. That means that when you have a given value of A260 for a solution of double-stranded DNA then when the DNA is completely denatured you will have a value of 1.36*A260. In your case you go from 1.00 to 1.37 so it means you have complete denaturation. This has nothing to do with degradation as was mentioned in the previous answer.

1) figure it out yourself - its just a bit of maths

2) the DNA becomes denatured at high temperatures and with extended exposure will degrade, thus causing your absorbance value to change
