There are several controls that could turn off the sound to the speakers. Go to Control Panel and find everything associated with "speakers" and "sounds" and "volume" and "sound card". I can't be more specific because I don't know your setup.
Naturally, make sure when you left click on the little loudspeaker icon in the toolbar that the slider is up. When you right click on that icon you should open up more sound controls.
Also, do you have a fancy keyboard with a volume control on it? Check that, too, then.
Do headphones work? If they do then you have a problem with the speakers, not the computer.

check things in this order:
1. The plug in for the speakers.
2. The volume knob on the speakers.
3. The speakers are plugged into the GREEN input on your PC.
4. The system Volume, there should be a speaker icon that you can double-click near the clock, if not, it is in your control panel, under the start menu. Make sure that no mute boxes are checked, and until you get everything going, turn all volumes to maximum. You can turn them down later.
5. The drivers for the sound card. This can most easily be done by going to start>control panel>maintenance>system>device manager. Look to see if there are any "!" inside a yellow block.

First of all, make sure your wiring IS right. Then, go into Control Panel, and make sure your sound system is visible to the computer. (You can also check Device Manager). Make sure you have the proper drivers (software) for your sound system. If your sound device is integrated into the motherboard, go into the BIOS and make sure that the device is activated. Look down on the tray (right hand side of the lower portion of your computer screen) and make sure you see a little horn, or sound icon there. If you do not, there's a software/configuration issue with your PC.

Go to your device manager and click on your sound device. You can try either updating for possible new drivers or uninstalling the the ones you have and they will reinstall upon restarting. Make sure all of your jacks and plugs are securely plugged in.

You may have a problem with the volume control. Other areas to look into is the final stage of your amplifier, see if you could find any burnt electrical components.
