I'd suggest that the second Amendment (right to keep and bear arms) is probably the most important from a practical point of view. Many Americans forget that the reason the nation exists is because groups of armed rebels fought against a governing body they believed was oppressive. The Declaration states that it is a civic duty to rise up and replace a government that becomes oppressive. The only right in the Bill of Rights that can guarantee the restoration of the others is the one allowing for an armed citizenry. Remember: Whether you believe the second Amendment gives a blanket "thumbs up" to gun ownership or not, it states that the reason to guarantee the right to keep and bear arms is that it is necessary to guarantee a free state.

Do you mean the most important Amendment? If that is what you mean...it's the 4th Amendment-Right to privacy. This Amendment has evolved into not so much guaranteeing the Right to Privacy of individuals, but to restrict power of law enforcement. Without it the good randomly search individuals without warrants, come into your house without reason and carry out racial profiling. Also, you could make a case for the 14th Amendment which requires the states to apply these Amendments to the citizens. This wasn't required until the 14th Amendment.
