Its really unlikely that spyware would advertise its presence by playing music, I think that the advice you've had to check the sound scheme is a good one for starters.
If that doesn't work post back, or try something like Ewido and an anti virus such as Avast!.

Go to the sounds and audio settings in the control panel. On the sound tab; set windows start sound to none.

Go to your start menu, and then click on control panel. then click on the sounds and multimedia icon. Then in the list of sound events go down to Start Windows. click on that, and then under name, there should be a drop down menu. go to the top of the menu and selece none.

I know of 800 OSes, that all play music upoon startup... Can't help you turn it off, if you can't tell us which OS you're running!
