IMHO, the ones that stick out in my mind the most are:
Pirates of the Caribbean: Curse of the Black Pearl - When Jack and Will are preparing to commandeer the Interceptor, the look that Sparrow gives when he turns and smiles just says "They don't stand a chance".
Jurassic Park - The T-Rex chase scene. That's just one of many memorable images from that movie.
Titanic - When the ship finally hit the iceberg. Personally, I didn't care about anything else about the movie until that point.
The Man in the Iron Mask - The ballroom scene when the Musketeers (sans D'Artangan) are messing with King Louis XIV by appearing and disappearing with the mask over their faces.
Star Trek II: The Wrath of Khan - The climatic final battle between the Enterprise and the Reliant.
That's all I can think of right now, but I'm sure there's more.

Best? Define "best".
Some great ones above that I had forgotten. But for me:
Sexiest? The Sundance Kid and Kate's first meeting in Butch Cassidy and the Sundance Kid.
Scariest? The first time I saw the "face hugger" grab John Hurt's face in the original Alien (I thought it was a space movie!).
Funniest? Probably the "Let me out of here!!!" scene from Young Frankenstein, where Gene Wilder has himself locked in with the monster, giving strict instructions that he not be let out, no matter what.
Most "Wow!"? Probably the opening 20 seconds of the first Star Wars (now called "Episode 4") the first time I saw it (on the huge screen at the Uptown Theater in Washington, D.C. on opening night!).
Best scene of all time? The one that I think of most that will be with me until I die? Can't name just one.

Western - Clint Eastwood's "Unforgiven" - Near the very end, the whole scene where he comes out of the saloon and he is yelling at the town to "bury need right." The first time I saw that scene, I thought he was going to kick my @$$!

Ok, considering my best, and not most known (known for me would be the burning of atlanta in GWTW), I'd have to say is when Bruce Willis realizes he's dead in sixth sense...shocked the you-know-what out of me. I love unpridictable movies, and that scene was cool.

Comedy: Office Space
In the beginning Micheal Bolton Is listening to gangster rap in his car during his work commute when a black guy selling pretzels (I think) walks by his car and Micheal sneaks is finger over and locks his car door. Funny! I love that movie! Sigh**
