Barefoot Gen and that was anime was based on actual events of the dropping of the atomic bombs on Hiroshima and Nagasaki in the eyes of a young man. One of the saddest moments when Gen's new baby brother died because the child starved to death.

I agree with both Ms. Sunlight and Muchi-Tsujo Kurai. Grave of the Fireflies and Bare Foot Gen are by far the most saddest anime. I would put GoF slightly ahead of Gen just because of the quality, but not so much, plus Gen has some light hearted moments that soften the blow. Both are wonderful, anime.

OK I change my answer I remember the saddest part I've seen is when Yusuke died in Yu Yu Hakusho. When his mother started to cry and then Keiko cries also it's so sad. It's even more sad when the little boy that Yusuke saved gave a greeting to Yusuke's body when he and his mother was going home he wanted to see Yusuke when he wakes up and he didn't even know Yusuke died saving his life and the little boys mom starts to cry. The old version of the tale about the female spider, I can't remember the title. But when she died at the end, but gave birth to hundreds of other baby spiders before she died had me in absolute floods of tears!!. I was only about 5 or 6 at the time!.

Fushigi Yugi was the only anime that made me want to cry more then once. When Miaka said good bye to Tomohome, when Soi died, when Hotohori died and when Nakego died.

blue on adult swim, it so sad the way the boy meet the girl the 1 time when they get isolated in a macgine but when he wake up from it other people and her was push up in a slime ball eye's open it........................so gross, but through the episode it so sad.
