i have a project on wheat karyotyping. we need to get good c-metaphase cells in order to karytoype it. our pretreatment is hydroxyquinoline. what is the best time to collect the roots tips, pretreat it with hydroxyquinoline and the duration of pretreatment. also, what is the best time to fix it in 3:1 abs. ethanol: glacial acetic acid? i know it is 12 pm for rice. how to hydrolyze it using HCl and stain with aceto-orcein. I know it's a lot of questions but i really need answers since we have been working it for a month now without acceptable results. thank you for your answers.
