The Yarra Valley is a great place to visit and taste some of Australia's best wines. You can generally do a day tour of the region (which is about an hour from Melbourne CBD) and cover about 6 wineries as well as lunch. If you want to experience a true aussie pub visit The Grand Hotel in Healesville. The public bar is often full of locals who are a great laugh if you chat to them.
A hot air balloon over Melbourne is a great way to see it and at the moment we have been having some fantastic weather.
If you like your sport visit the MCG while the AFL football is on as it is a great Aussie sport you won't see played professionally anywhere else in the world. We just had the Commonwealth Games in March so there is plenty to see in regards to that sort of thing too.
When you arrive in Melbourne, go to the Visitor Centre on the corner of Flinders and Swanston Sts at Federation Square. They have plenty to offer, even free tour guides that will show you things you want to see - shops, architecture, fashion, whatever you like!

I think its all been covered but add gippsland to your list and visit the ski fields around Mt Baw Baw aswell.
and suburb wise stay away from St Kilda Road, you may get mugged or raped.
go to Sprigvale, but if you have time goto Phillip Island and see the penquin parade-its something that cant be passed up.
