The Gräfenberg spot, or G-spot, is a small area in the genital area of women behind the pubic bone and surrounding the urethra.
Every woman is shaped differently on the inside so some women have a harder time finding their G-spot than others. Likewise, some women are not sensitive in that area so it won't have the same affect on everyone (if you hit it during intercourse or otherwise).
Typically, the best way to find it is to place your index finger (palm up) inside your vagina and then make a 'come here' motion with it. Your G-Spot should be right over that little 'ledge' that you'll feel.

It varies.
Unlike the clitoris, many women do not have a "G-Spot"; its not an anatomical location recognized by any doctor or university.
That said, many people claim that its a few inches "in" and on the top side of the vaginal canal, if the woman is on her back.
Women that claim to have a G-spot rarely agree on its exact location as well. It basically boils down to finding the spot that feels good for that particular woman.

It was once believed to be a bundle of nerves in or around the vaginal walls but is now reported to be the same as, or part of, the urethral sponge (Heath, 1984), the site of Skene's glands, a homologue of the prostate that exists in many women believed to be the source of female ejaculation. It is located on the ventral side of the vagina, about half way between the pubic bone and cervix. When this spot is stroked, there is a sensation or urge to urinate, but if the stroking is continued during sexual arousal it can be sexually pleasurable (Shibley Hyde, J. and DeLamater, J.D., Understanding Human Sexuality, Eighth Edition (2003)). For some women, it can be a primary source of stimulation leading to orgasm during intercourse while having sex in positions that use the penis to stimulate the frontal wall of the vagina.
The G-spot may not be just one discrete spot. Natalie Angier contends that it is merely the deep nerves of the clitoris as they pass through the tissue to connect with the spinal column. The clitoris has deep roots and may in fact change in size and slightly change in location as hormone levels fluctuate throughout a woman's life.

About and inch or so inside her vagina on the upper wall under her pelvis. You can sometimes feel it with your finger - like a little rough spot, and it can swell a little when she's excited.
It's been rumored to also exist in your wallet.

the 'G-spot' is located 2 inches up on the front wall of the vagina...normally it is about the size of a pea..but upon being stimulated and it arrouses the female, it can grow upwards to about the size of a walnut.when you stick your fingers in her and do the 'come here' motion, if she squirms or moans, you hit the spot.
