It depends on what you're trying to make a shortcut to.

Programs - find the program in your start menu and right click on it, copy it, and paste it onto your desktop.

Sites - click the button inbetween the minimize [_] and close [X] buttons to make it so you can see a bit of your desktop and the browser at the same time.

See the address bar in your browser? See the little icon at the beginning of it? Drag and drop that onto your desktop.

If the program is already installed, go to Start and right click it > then go to PROGRAM FILES and double click it > then scroll down till you see a folder named YAHOO and double click it > Then go to the folder that is called In that folder is a file called Yahoo Shortcut. Copy and paste that to your desktop & quick launch bar And there is a short cut with a quick launch to boot!

Right click your desktop, point to new, go to shortcut. Click browse, then find where the file is you want to make a shortcut to. Alternatively, right click the file you want to make a shortcut of, and click make shortcut, then drag it to your desktop.
