"It's always Darkest before the Dawn" is an old proverb. It tells us that in life sometimes we have to go through the darkest times before we can see the light, the Dawn.
AND it has been proved that it IS NOT "literally" the darkest before the dawn. I would deduce that the darkest hour would be when the sun is the furthest from the Earth. Which is approximately December during the winter solstice around 1am.
"The Darkest Hour" is a coined phrase from Winston Churchill describing the time during WWII when Great Briton was in danger of invasion.

There are two answers to this question:
The human version is loss of sight of one's purpose in the whole, or a separation from the creative force in the universe (aka god). This is seen in cancer cells, suicidal maniacs, and depressed folks. The opposite is seen in astronauts and spiritual (not nessesarily religious) leaders, old wise folks, new moms, etc.
The daily earth version would be the half way point between the time of sunset and sunrise for the day you want to observe it. So if the sun sets at 8pm and rises at 8am, the halfway point would be 2am. You can find the sunrise and sunset times in your local online weather or farmer's almanac.

First of all darkness is a relative term. Depending on where you are located and how much light pollution there is in your area determines how much darkness is possible in your area. Midnight using standard time (not daylight saving) on the night of the new moon. That way, you get no moonshine.
