When defragemting the hard drive, it won't make any difference how many users you have on that PC. The Defrag program will "optimize" the entire hard drive.
I am assuming that you are using the Microsoft Defrag program when asking this question. It works the same way for Norton's Speed Disk too.

If your computer is like most people's, mostly unused disk space, you can go years without having to defragment. But you will get better performance from your disk if you defragment every month or so.

Only if you are running Windows 9x or Windows XP. OSX does not require it and neither does Linux to my knowledge. I do it every two weeks, if you don't use your computer much to create files and delete files or uninstall and install a lot of programs then you can get by with doing it maybe once a month.
