I have a friend who is very funny and friendly and happy when he drinks but withdrawn and glum and boring when he is sober. He says he wishes he could always be the way he is when he's had a few drinks, because that is who he really is on the inside, but for some reason he can't get over his inhibitions.

what a good question, well you don't need alcohol or drugs, you need to be your self and empower them, encourage, build their confidence perhaps by:
ACCEPTANCE AND POSITIVE REGARD
listening to where they are at, not putting your own journey on some one, this will encourage them to explore all possibilities, they may have different experiences from your self and different values, you need to listen and be positive to encourage change.
EXCHANGE OF SELF AND OTHER
When the good things we offer are not received by others, we are inclined to despair, appreciate the viewpoints of this person and then share your experiences, let them see that they are liked and respected, recognise what is holding them in this place..If they are using alcohol or drugs to self medicate, given them confidence, you need for first understand the reasons why and then reflect back to them some positives..
sorry this is heavy but it depends on why some one finds it hard to be more spontaneous or has these inhibitions to start with.. show them your skills and interests, do it gently and with encouragement.

IN order for a person who has been drinking for a resonably large amount of time you need to get them to be sober for long enough to get the alcohol out of their system so they can see what they are missing in life .

Depends. What are YOUR motives towards this person?

If you just like them as a person, then just be yourself. If they like you for you, then they will relax naturally on their own so long as there's no pressure from you or the environment.
