An excellent program for doing this is Tune-Up Utilities 2006 which will perform cleaningn tasks and also registry improvements. For spyware look to specialist programs like AdAware, etc. Go to www.download.com and get several test programs and find what works for you.
TUU2006 is the bee's bits though in terms of improving general speed in my experience and even works well on servers.
