A proxy is a computer that gets internet requests on behalf of your computer. In other words say you are looking for www.yahoo.com. your computer makes a request out to the proxy and the proxy gets the webpage and sends it to your computer. Normally you don't need to worry about proxies because most computers have an shared connection through a cable/dsl router or their own modem. It is were people don't have this luxery and are allowed only one computer that can access the internet and want to share it with others within their network without a router as described above. It is also common to see spam / popup blockers using proxies to filter out inappropriate content or keep a list of frequently accessed pages so that they can be retreived faster in future requests.

I'm assumming that you're running Internet Explorer and I'll go through those steps. You can either right-click the Internet Explorer icon and click properties or open Internet Explorer and click on tools menu and go to internet options. It should bring up a new window with several tabs and go to the connections tab. At the bottom is lan settings which will give you access to change proxy settings. Normally on this page Automatically detect settings is activated, but it will usually work fine if its disabled. Depending on what softwares you have on your computer or network settings, you may have Use automatic configuration script or proxy settings enabled. I wouldn't change these unless you know what you are doing. This is because you may have a spam or popup blocker that needs these settings or your ISP or network admin if you're at work needs these settings there to make your computer to work properly.
