Take it to a jeweler if it's in a setting. They can do a quick and free test on it. However, some modern stones can fake out the testers. Get a reputable one to look at it for you. They can look at it under their tools to tell.
If it's in a cheap setting, it's fake. If it's in an expensive setting, chances are it's real. Look for a stamp on it. On a ring, it's on the inside of the band. On a bracelet or necklace, it's on the clasp. If it's earrings, look at the back. If it doesn't have a stamp, it's fake.

For a loose stone:

Good question... ask a jeweler is your best bet. CZ are perfect... diamonds are not and cost will be your biggest indicator. A "perfect" diamond is ridiculously high in price where a CZ wil be $100. When you peer into the diamond, you will see little black charcoal or black splotches... about the size of a pin head depending on the "clarity" of the diamond. Try diamond.com to read up on the 3C's and how to pick the right one. This will at least give you some background on diamonds!

BE CAREFUL AND SHOP FROM A REPUTABLE STORE!!!

Jewellers use high magnification lenses and specialised electronic equipment to tell diamonds from fakes, like cubic zirconia or moissanite. It's not possible to tell with the naked eye alone if something is definitely a diamond, although a trained eye can sometimes tell if something is definitely not a diamond with large stones.

Synthetic stone companies are working to improve their products all the time. If you need to know, go see a pro.

I'm curious. Oh, I do know one thing; they sparkle with rainbow colors. But it would take a pro to be sure.
