Marcelles Wallace's soul.
Take special note of the suitcase's lock combination: it was 666, that being the sign of the beast/devil.
The 2 hitmen recovered the suitcase and were seen defending it with their lives to get it back to Wallace, who presumably sold his soul to the devil to attain wealth and power. The hitmen then, can be viewed symbolically as fallen angels in Wallace's service, returning his lost soul to attain redemption in God's eyes.
Note too, in Samuel L Jackson's speech, he refers to himself as 'the finder of lost children.' Wallace, a criminal mastermind/kingpin, can be viewed as the ultimate lost child - the suitcase containing a myriad of other things such as innocence, purity (gold color), piety etc...getting the suitcase rewards wallace with redemption and forgiveness, tying his existence and mission inexorably to the successes/failures of the hitmen.

It was rumored to be Marcellus Wallace's soul. Hence the combination being 666. It was never revealed what was actually in the suitcase though.
In a radio interview with Howard Stern in late 2003, Quentin Tarantino was asked by a caller the contents of the briefcase, and he answered, "It's whatever the viewer wants it to be."

I always thought it was money but maybe it was gold. The movie was such a black comedy that it could have been nothing more than a suitcase full of Quarter Pounder's Deluxe.....

They never say.
And they dont really need to. The whole point of the suitcase is to drive the plot, it really doesnt matter whats inside it as long as it is something we assume is valuable.

It was never said or shown, he says its his boss's 'dirty laundry' but of course it isnt. It appears to be a reflection of gold coming from the case, but the audience is never shown. I dont think it is Marcelis soul bc the guy robbing the coffee shop at the end see it & is amazed by its value/contents, why would he give a shyt about it, if it was somebody's soul?
