Apparently, the most isolated capital city in the world is Perth, Australia
Perth is over 4400km (2750mi) from Sydney by road. I'm sure you would find more remote locations, but maybe they're not cities.

If you mean actual city, not too sure, but I do know the Town of Nunavut in the Northwest Territories of Canada is around 400 miles to the nearest town of any size.....
