well Lake Superior is the largest lake, but it is not entirely in the US, it's northern border is Canada. Lake Michigan is the largest US lake entirely within US borders.
How long would it take to get there on a boat from......where? You give no starting point....From where I am it would take forever, I wouldn't go there by boat, I would drive....

I believe the answer you're looking for is Lake Superior - the answer changes depending on if you're speaking in terms of area or volume - as far as how long it takes to get there, depends on where you're coming from....but, if you take that bus, you'll get there.
