Please go see the Orchid Gardens. I have been all over the world, and that is one of the most spectacular sights I have ever seen... so many acres of so many different varieties of exquisite orchid blossoms that you just can't even take it all in. If you have any interest in photography at all, this place is a treasure.
No visit to Singapore is complete without a visit to Raffles for a Singapore Sling in the Long Bar. Also, the India Town section of the city is a cultural experience and has good food and shopping. The Chinatown area is worth about 15 minutes - it has been too sanitized to have any real cultural interest any more. But if you are a history buff, Changi Prison (of King Rat fame, it is still there) is an experience of a completely different kind.
Just try to avoid Orchard Road if at all possible. It has to be the absolute worst tourist trap on the planet. I was there in late December, and there were more tinsel christmas trees and santa clauses on Orchard Road in Singapore than in a whole century's worth of Macy's parades... yuck.

You can visit the Night Safari, the Zoo, Jurong Bird Park, Downtown East in Pasir Ris, Chinatown, Little India, Sentosa or if you are the clubbing kind of people, can always visit the pubs along Singapore river. For food, you can go to Newton Circus Food Centre. If you want to watch theatrical works, then hop over to the Esplanade. Or visit the farms in Singapore.

I went to the zoo and night safari the other day and it was awesome. I also hear good things about something called stone steak which you can get near the quays. I may go hunting for it in the week.

Kite flying, rollerblading, fishing, prawn fishing, checking out the science centre (omg when's the last time i've been there?!), you can go check out the museums too.
If you prefer the more city-kinda activities... catch a movie? go play a round of billiard or pool... go for karaoke.. just to name a few :)
