Nikko is definitely the best of the 2 options but if its's just one day, you need to get up really early. It'll take 2/3 hours to get there depending where you are staying in Tokyo. Also, everything will close at 4:30-5:00pm
If you are a late sleeper of don't fancy the train journey then head to Kamakura. It'll take about an hour to get there, and there's still plenty of temples and the Buddah to keep you occupied for a day. Also loads of little gift shops for souveniers to take back home. If you have enough time, take a ride on the quaint Enoden line to Enoshima, a pleasant island just along the coast. On the way back to Tokyo, stop off at Yokohama and head to Sakuragicho - Minato Mirai. An unrivaled night view. Including Japan's tallest building and the world's biggest clock (on a ferris wheel).

Never been to Kamakura, but I can share with you what you can see at Nikko.
The main attraction would be the waterfall. Honestly, it's just a waterfall, nothing spectacular. However, I love the lake, with lots of typical Jappy shop-houses and eateries around it. Very nice place to stroll and take pictures. My hubby and I took this tour bus from Tokyo, the sceneries were really quite interesting.. gives you all the Jappy feeling.

I definately would recommend Kamakura, go see the Great Budda ... Nikko is pretty, but can be very boring.

You can see lots of old temples and shrines in Kamakura, they are beautiful but you may get bored. Rahter you better go to Nikko and see one very beautiful temple, "日光東照宮," a very impressive one, really.

I would go to Nikko in that situation. The giant Buddha is nice, but that can't really fill a full day.
