I'd say the first science fiction film would be Georges Melies "La Voyage dans la Lune" ("Voyage to the Moon") in 1902.
The first outstanding science fiction film--outstanding in that it influenced later films--would probably be "Metropolis," in 1926.
"King Kong" in 1933 was probably the first sci-fi movie that appealed to a mainstream audience.
"2001: A Space Odyssey," in 1968, is usually considered the first science fiction film that had a deep, philosophical level to it. It is usually considered one of the greatest films ever, and it used realistic special effects that are impressive even today.
"Star Wars: A New Hope," in 1977, is probably the first movie that could be called "a blockbuster"--science fiction or otherwise.

actually the first non documentury film ever was a science fiction movie: it was made by Melies (inventor of cinema) in France around 1900 and was called 'Voyage to the Moon'. It's about 10 minutes long and has special effects of sorts (flying rocket, extra terrestrials) but it's really just a spoof based on Jules Vernes novel of the same name.
Metropolis by Fritz Lang is probably the first feature length one though.

I don't know what is actually the first one, but the oldest I've seen is called "Metropolis" from 1904, by a guy named Fritz Lang or something...
There is an anime remake of it that they did about five or so years ago, but the original is pretty old school. I didn't even know they had movies back then, but some of the sets for Metropolis are huuuuge!
